It has been almost twenty‐five years since Huberman and Riggs first showed that there are multiple bidirectional origins of replication scattered at ∼100 kb intervals along mammalian chromosomal fibers. Since that time, every conceivable physical property unique to replicating DNA has been taken advantage of to determine whether origins of replication are defined sequence elements, as they are in microorganisms. The most thoroughly studied mammalian locus to date is the dihydrofolate reductase domain of Chinese hamster cells, which will be used (...) as a model to discuss the various methods of investigation. While several laboratories agree on the rough location of the [initiation locus] in this large chromosomal domain, different experimental approaches paint different pictures of the mechanism by which initiation occurs. However, a variety of new techniques and synchronizing agents promises to clarify the picture for this particular locus, and to provide the means for identifying and isolating other origins of replication for comparison. Mayhew claims that the objections have been largely ignored on the ground that they are facile or unfair. But the purpose of the book is not to show that Aristotle’s thought has been unjustifiably vilified, though (...) Mayhew says that “we shall see in passing that in most cases Aristotle’s criticisms of the Republic are well-founded”. The book’s purpose is to see how the objections illuminate the rest of Aristotle’s political thought. I argue that desire satisfaction theories of welfare are not committed to the view that changes in welfare levels can happen after death, or that events that occur after death impact the agent's welfare levels now. My argument is that events that occur after death have only epistemological import. They may reveal that the person was successful (unsuccessful) in life, but the desire was already frustrated or satisfied before the person died. The virtue of the account is that it gives (...) us a way to acknowledge both the intuition that we cannot be harmed after we die and, in a sense, the intuition that things that happen after we die are relevant to our lives. The child was 2 years, 8 months old and weighed 25 pounds, one-fifth the weight of her mother, for whom she was to be the bone marrow donor. The mother had suffered a relapse of acute myelogenous leukemia; her physicians recommended a bone marrow transplant. The child was the closest human leukocyte antigen match and thus the best donor candidate for her mother's transplant.

We highlight the various domains of the flavivirus virulence factor NS1 and speculate on potential implications of the NS1 3D structure in understanding its role in flavivirus pathogenesis. Flavivirus non‐structural protein 1 (NS1) is a virulence factor with dual functions in genome replication and immune evasion. Crystal structures of NS1, combined with reconstructions from electron microscopy (EM), provide insight into the architecture of dimeric NS1 on cell membranes and the assembly of a secreted hexameric NS1‐lipid complex found in patient sera. (...) Three structural domains of NS1 likely have distinct roles in membrane association, replication complex assembly, and immune system avoidance. A conserved hydrophobic inner face is sequestered either on the membrane or in the interior of the secreted hexamer and contains regions implicated in viral replication. The exposed variable outer face is presented to cellular and secreted components of the immune system in infected patients and contains candidate regions for immune system modulation. We anticipate that knowledge of the distinct NS1 domains and assembly will lead to advances in elucidating virus‐host interactions mediated through NS1 and in dissecting the role of NS1 in viral genome replication. For many firms, a focus on corporate social responsibility (CSR) is an indication to stakeholders that the firm is concerned about social and environmental issues. However, these same firms may engage in CSR activities with the expectation that these activities will increase their bottom line. A relevant, and highly researched question, is the relationship between CSR and performance. The findings are inconclusive, indicating a need to consider other explanations. Several authors have drawn on the resource-based view of the firm to (...) suggest that CSR can give a firm a competitive advantage. In this article we draw on and further develop this research, by focusing on four outcomes of CSR initiatives: strategic disadvantage, strategic necessity, temporary strategic advantage, and strategic advantage. We exemplify this with two cases. The article contributes to the literature by developing a set of possible outcomes, by recognizing several issues which result in strategic necessity, and by suggesting movement between the outcomes. Elephants show a rich social organization and display a number of unusual traits. In this paper, we analyse reports collected over a thirty-five year period, describing behaviour that has the potential to reveal signs of empathic understanding. These include coalition formation, the offering of protection and comfort to others, retrieving and 'babysitting' calves, aiding individuals that would otherwise have difficulty in moving, and removing foreign objects attached to others. These records demonstrate that an elephant is capable of diagnosing animacy and (...) goal directedness, and is able to understand the physical competence, emotional state and intentions of others, when they differ from its own. We argue that an empathic understanding of others is the simplest explanation of these abilities, and discuss reasons why elephants appear to show empathy more than other non-primate species. To hold an error theory about morality is to endorse a kind of radical moral skepticism—a skepticism analogous to atheism in the religious domain. The atheist thinks that religious utterances, such as “God loves you,” really are truth-evaluable assertions (as opposed to being veiled commands or expressions of hope, etc.), but that the world just doesn’t contain the items (e.g., God) necessary to render such assertions true. Similarly, the moral error theorist maintains that moral judgments are truth-evaluable assertions (thus contrasting (...) with the noncognitivist), but that the world doesn’t contain the properties (e.g., moral goodness, evil, moral obligation) needed to render moral judgments true. In other words, moral discourse aims at the truth but systematically fails to secure it. If there is no such property as moral wrongness, for example, then no judgment of the form “X is morally wrong” will be true (where “X” denotes an actual action or state of affairs). Advocates of this position include Hinckfuss 1987; Joyce 2001; Mackie 1977 (see MACKIE, J. L.). Various forms of moral skepticism—some of which are arguably instances of the error theoretic stance—have been familiar to philosophers since ancient times. (See SKEPTICISM, MORAL.) Error theoretic views can be controversial—as in the case of religion and morality—or widely agreed upon—as in the case of ghosts and phlogiston. It is important to note that error theorists maintain that the judgments in question are erroneous not merely because of the absence of any objective moral facts sufficient to render them true, but also because of the absence of any non-objective moral facts sufficient to render them true. There is, for example, a kind of moral realist who maintains that moral properties are objective features of the universe (see REALISM, MORAL). JenkinsIt is common to open a work regarding the merits of Mansfield Park by noting that Fanny Price is very difficult to like. Nietzsche might have described her as a "moral tarantula." 1 She sits, making negative moral judgments about the actions of others, while doing nothing herself. Fanny spends most of her time, literally, sitting or lying down. Austen describes her character (...) as "supine." Comparing Fanny with her sister Susan, Austen says that "Susan was only acting on the same truths, and pursuing the same system, which her [Fanny's] own judgment acknowledged, but which her more supine and yielding temper would have shrunk from asserting. Susan tried to be useful, where she could only have gone away and cried" (MP, p. 395). 2 And Fanny cries all of the time. She cries at least thirteen times over the course of the novel. So, there are two questions about the novel that need to be answered. Why did Austen create a heroine who is so clearly distasteful? Why do some critics and philosophers see Fanny as a moral ideal or cultural icon? Some (Alasdair MacIntyre, Gilbert Ryle, Lionel Trilling) argue that Fanny is so unappealing just because Austen wants to be didactic 3 and force us to focus on virtue. According to Lionel Trilling, for example, Mansfield Park has a different moral message from Pride and Prejudice. "Its praise is not for social freedom, but for social stasis. It takes full notice of spiritedness, vivacity, celerity and lightness, but only to reject them as having nothing to do with virtue and happiness, as being, indeed, deterrents to the good life." 4 Others think that the infelicities of the novel and its heroine make it a parody of traditional feminine values and conservative moral thought in general. 5 Here, I shall argue that a path between these two extreme ways of reading the novel is correct. I shall argue, using Alasdair MacIntyre's discussion as a starting [End Page 346] point, that Austen does not cast Fanny as a moral ideal. If Austen is Aristotelian (a common view among philosophers) Fanny cannot be an ideal. And, although I agree with Johnson's view that those who have read the novel as morally conservative are wrong, I do not think that the novel exhibits global skepticism about traditional standards for the beautiful/feminine. Austen condemns the idea that women should be passive dolls who do not think for themselves, but she does not parody the traditional idea that women should be sweet, gentle, and caring. In my view, Fanny's character constitutes a criticism of passivity, and of rigid adherence to moral rules.Alasdair MacIntyre's view is that Austen is an Aristotelian moralist with a Christian slant on the make-up of the virtues. He describes Fanny's character as an ideal of virtue. He argues that Fanny is pretty much lacking in charm just because Austen wanted to focus on the nature of virtue, which charm can obscure and which Fanny is meant to epitomize.For charm is the characteristically modern quality which those who lack or simulate the virtues use to get by in situations of characteristically modern social life... And the charm of an Elizabeth Bennett... may mislead us... Fanny is charmless; she has only the virtues, the genuine virtues to protect her... and when she disobeys her guardian, Sir Thomas Bertram, and refuses marriage to Henry Crawford it can only be because of what constancy requires. It also addresses the dynamics at the meeting point of multidisciplinary practice-led design research.

In 2010, pitcher Armando Galarraga was denied a perfect game when umpire Jim Joyce called Jason Donald safe at first with two outs in the bottom of the 9th. In the numerous media discussions that followed, Joyce’s ‘blown’ call was commonly referred to as ‘mistaken’, ‘wrong’, or otherwise erroneous. However, this use of language makes some not uncontroversial ontological assumptions. It claims that the fact that a runner is safe or out has nothing to do with the ruling (...) of the umpire himself, but rather with some state of the universe that does not depend on the umpire for its existence. In this paper, I recast the problem as a version of Plato’s Euthyphro Dilemma and argue that the view implied by the above assertions is actually misguided. Instead, I hope to show that an alternative view – what I call ‘restricted umpire voluntarism’ – is actually more in line with the spirit of the game of baseball and is not as counterintuitive as it may appear.. In the twentieth century, most PhD-trained academic philosophers in both the United States and United Kingdom were white men. The first black woman to earn a PhD in Philosophy was Joyce E. Mitchell Cook. A preacher's daughter from a small town in western Pennsylvania, Cook earned a BA from Bryn Mawr College. She went on to earn degrees in Psychology, Philosophy and Physiology from St Hilda's College at Oxford University before earning a PhD in Philosophy from Yale University in (...) 1965. At Yale she served as Managing Editor of the Review of Metaphysics and was the first woman appointed as a teaching assistant in Philosophy. She taught at Howard University for nearly a decade and held positions in national government service in Washington, DC, before retiring to a life of independent study of the black experience. Although she did not publish much in her lifetime, Cook deserves to be remembered as: first, an academic trailblazer who proved that race and gender are not barriers to excellence in philosophy; second, a public philosopher who broke barriers as a foreign and economic affairs analyst and presidential speech writer; third, among the first philosophical bioethicists of informed consent and experimentation on humans; and, fourth, an analytic philosopher of race, opposing claims that blacks suffer from inherited intellectual inferiority. Cook's achievements can inspire women of all backgrounds who love philosophy to pursue graduate studies and academic careers. (shrink)

Walkowitz (bio)For the first seven years of my career, I taught a very large lecture course once, and sometimes twice, a year in a graded auditorium filled seat-to-seat with as many as 350 undergraduates. The course focused on a cluster of themes that linked art and violence–how art resists violence, how art animates violence, how art expresses violence, how violence spurs art– and traced those themes (...) through the history of British fiction from Joyce, Woolf, and Forster through Barker, Burgess, Ishiguro, and Rushdie. I loved teaching that class. I loved and was sometimes terrified and almost always invigorated by being in that room with those students. Needless to say, no one wore masks. Far from it.I remember the feel of the crowded, anonymous, yet intimate room. Intimate, because a group of 20 or 30 very avid students occupied the first few rows, and I came to value and recognize their attentive faces. I could hear their little sounds as they scratched out notes, adjusted their laptops, whispered to each other, and rustled papers. Intimate, because I was standing at my full, not very substantial height on a stage, and they were looking up at my body and listening to my voice for 50 minutes twice a week. And intimate, in another sense, because they would sometimes forget that we were there together live and behave in ways that I have never experienced in a smaller classroom. One day, two students in the last row, which in the slanted auditorium was directly level with my elevated gaze, spent most of the lecture kissing ardently. I was impressed by their enthusiasm and by the duration of their embrace. At the end of the lecture, I quick-stepped up the aisle to remind them: this is not TV; we are here together. I smiled while I said it, and they apologized for distracting me. I smile as I think about it now.During the COVID lockdown, I missed the embodied experience of large gatherings, including large lectures. The Zoom classroom, for all its affordances, can't replicate the accidental encounters, the risk and energy of embodied presentation, the warmth of individual eye contact, and the shoulder experiences of the class period: what happens in the time just before and the time just after the actual class takes place. The shoulder experiences are truly precious, as any seasoned teacher knows, because [End Page 258] they allow us to shift the scale of encounter up and down, and they allow us to interact in spontaneous and less formal ways than we do when the entire class is gathered as one. Close up and informally, we are more likely to use our faces and our hands: smiling, nodding, tilting, waving, and pressing our palms together to say thank you.There is a kind of social intimacy and a kind of social violence that can only be illustrated in a very large group, and I appreciated my lecture for allowing me to experience that. The most terrifying day of my class each semester was the day I spoke about– and performed!–the shock of involuntary, inchoate expression in E. M. Forster's A Room with a View. Like other Bloomsbury artists, Forster was committed to ushering out traditional ideas of self-restraint and maturity and ushering in greater communication between men and women, between social classes, and between ages about sexuality, desire, and the experiences of the body. The word of the day was "play": life needed more play, more honesty, more curiosity, and less seriousness, less decorum, and less lying to ourselves and others.What does this look like in the classroom? On the day in question, we would talk about the scenes in the novel in which characters are criticized for disruptive actions they cannot or do not control, such as fainting, laughing, bleeding, getting carried away, or expressing sounds in addition to words. I would explain that there are two problems with these actions: they are accidental or unintentional, and they involve an unexpected, unregulated expression of the body in public.
